Weizmann Institute Researchers Ayelet Erez and Yarden Ariav Reveal How Sildenafil May Restrict Cancer Metastasis

Researchers at the Weizmann Institute of Science have made a discovery poised to transform approaches to oncological therapy. They found that sildenafil—the active ingredient in Viagra—can restrict metastasis. This offers an entirely new perspective on how existing medications could prevent the spread of malignant cells throughout the body.
A team led by Professor Ayelet Erez and Dr. Yarden Ariav closely examined how cancer cells detach from the primary tumor and migrate to healthy organs. They established that to successfully relocate and adapt to a new microenvironment, cancer cells critically require vast amounts of cholesterol. This lipid serves as an essential building block for forming elastic and resilient cell membranes, without which the migration of malignant cells is impossible.
During their experiments, the biologists discovered that sildenafil effectively disrupts the internal logistics of the cancer cell. The drug inhibits a specific enzyme, PDE5, which inevitably leads to a sharp increase in the levels of the signaling molecule cGMP. This molecule binds to the proteins responsible for intracellular cholesterol transport, literally cutting off access to this vital resource. Deprived of cholesterol, the cell loses its ability to construct the membranes necessary for metastasis and remains trapped within the primary tumor.
The study’s authors see enormous clinical potential in this newly identified mechanism. They posit that, in the foreseeable future, oncology could adopt a combination therapy pairing sildenafil with standard statins—drugs traditionally prescribed to lower overall blood cholesterol levels. This dual strike- targeting both the body’s systemic cholesterol volume and its transport within malignant cells-could become a highly effective and accessible tool in the fight against aggressive forms of cancer.
